Konami offers players the chance to once again strap on a guitar and become a legend. Over 35 new songs have been added including both popular Japanese and western songs. This version also includes NONSTOP mode which covers the ORDER, OFFICIAL and RANDOM courses. The more popular BONUS TRACK has its own song.

The "Multisession Mode" seen in GuitarFreaks 5th Mix also makes a return. In this mode, players create a super jam session by linking with Keyboardmaina 3rd mix and Drum Mania 4th mix, with up to 5 players able to play together simultaneously (two on Keyboardmania 3rd Mix, two on Guitar Freaks 6th Mix, and one on Drummania 5th Mix). When linked with Keyboardmania, about a dozen songs are available to play.

Gameplay remains the same from previous titles, with the player using a simulated guitar controller to play the notes of a chosen songs guitar part as they are displayed on the screen.
