Metal Slug 6 is the first installment of the series developed on the Atomiswave arcade hardware system. The number of playable characters has expanded for the first time since Metal Slug 2, with 6 characters available for play. Newcomers Ralf Jones and Clark Steel of King of Fighters and Ikari Warriors fame join the original team of Marco Rossi, Tarma Roving, Eri Kasamoto and Fio Germi.
There are two difficulty levels, easy and hard, and the new weapon stock system allows players to carry two gun power-ups at the same time. Two new player control buttons have been added to allow for greater control over the action, and two new vehicles have been added.
